The mechanisms of formation of secondary immunodeficiency in action acrylonitrile
Acrylonitrile (AN) , widely used in chemical industry, can lead to acute, subacute and chronic intoxications accompanied postintoxication immunodeficiency. The aim of this study was to investigate the mechanisms of the action of secondary immunodeficiency acrylonitrile. In experiments on albino rats weighing 180-240 g were administered subcutaneously AN . The acute intoxication in doses of AN - 0,25; 0,50 and 0,75 LD50 - is the dose-dependent reduction in the number of colony forming units (CFU ) in the spleens , respectively, in 1,5; 2,2 and 3,3 times (p< 0.05). In subacute ( for 6 days ) and chronic effect of AN 30 and 60 days , respectively, at doses of 0,01 and 0,005 LD50 CFU marked reduction of migration from the bone marrow to the spleen in 2,5; 2,1 and 2.3 times (p<0.05). The AN
suppresses migration of bone marrow B cells and T lymphocytes precursors . After
the ex vivo steps AN after 1 day B cells function in cooperation with T cells decreased by 1,4 times (p<0.05), and T-cell function - 2,0 by 1,4 times (p< 0.05) compared with control. The AN to 0,75 LD50 dose after 3 days decreased the activity of acetylcholinesterase of T cells and splenocytes, thymocytes , respectively, in 1,4 and 1,5 times (p< 0.05). A similar effect was observed in subacute and chronic action of the AN. The toxicant dose-dependently decreased the content of ?-
naphthyl- AS -atsetatesterase and ?-naphthyl-butiratestesterase in T lymphocytes, monocytes, macrophages of splenocytes. An expression line (r = 0,69-0,78)
correlation between the parameters of the immune system and indicators of antioxidant system (catalase, peroxidase) and reverse
(r = - 0,67-0,78) correlation between the parameters of the immune system and indices of lipid peroxidation (malondialdehyde , total production of radicals).
